Overview

Immunoglobulin E (IgE) antibodies specific for Der f 2 are a specialized subset of antibodies that recognize the major allergen Der f 2, a 14-kDa protein from the house dust mite Dermatophagoides farinae (Thomas et al., 2010). These antibodies are central to the Type I hypersensitivity reactions that drive house dust mite-associated diseases, including allergic asthma, perennial rhinitis, and atopic dermatitis (Galli & Tsai, 2012). In sensitized individuals, Der f 2-specific IgE binds to high-affinity FcεRI receptors on the surface of mast cells and basophils. Upon subsequent exposure to the allergen, the IgE molecules are cross-linked, triggering the degranulation of these cells and the release of potent inflammatory mediators such as histamine, leukotrienes, and cytokines. Therapeutic interventions targeting this pathway include the monoclonal antibody omalizumab, which sequesters circulating IgE to prevent its binding to receptors, and allergen-specific immunotherapy (AIT), which utilizes Der f 2 extracts to induce immunological tolerance and reduce the production of specific IgE (Normansell et al., 2014). Measurement of serum levels of these specific antibodies serves as a primary diagnostic biomarker for identifying mite-sensitized patients and monitoring treatment efficacy.
