Immunoglobulin E (IgE) antibodies specific for Glycyphagus domesticus allergens are the primary mediators of allergic sensitization and Type I hypersensitivity reactions to this storage mite (Gafvelin et al., 2001, PMID: 11531051). Glycyphagus domesticus, commonly known as the grocer's itch mite, is a significant source of indoor and occupational allergens, particularly in damp environments or agricultural settings (Armentia et al., 1997, PMID: 9194931). These patient-derived IgE antibodies recognize and bind to specific mite proteins, most notably the major allergen Gly d 2, which belongs to the NPC2 protein family (UniProt P81528). Upon binding, the IgE-allergen complex cross-links high-affinity FcεRI receptors on the surface of mast cells and basophils, triggering the degranulation and release of pro-inflammatory mediators such as histamine and leukotrienes (StatPearls, Type I Hypersensitivity). This physiological response results in clinical manifestations including allergic rhinitis, bronchial asthma, and dermatitis. Therapeutic intervention typically involves the use of anti-IgE monoclonal antibodies, such as omalizumab (Xolair), which bind to the Fc portion of circulating IgE, thereby preventing its attachment to effector cells and downregulating receptor expression (FDA, Xolair Prescribing Information).
Omalizumab binds to the Cε3 domain of the IgE heavy chain, preventing the interaction of free IgE with the high-affinity FcεRI receptor on mast cells and basophils, thereby inhibiting the allergic cascade.
