Immunoglobulin G antibody against SARS-CoV-2 spike protein or nucleocapsid protein (IgG against SARS-CoV-2 S/N protein)

Target
IgG against SARS-CoV-2 S/N protein
Molecular classification
Immunoglobulin (antibody), Other (biomarker/generic immune effector)
01

Overview

Immunoglobulin G (IgG) antibodies against the SARS-CoV-2 spike protein or nucleocapsid protein develop as part of the adaptive immune response after infection with SARS-CoV-2 or vaccination. Anti-spike IgG can neutralize the virus by blocking its interaction with the host ACE2 receptor, while anti-nucleocapsid IgG reflects prior viral exposure but is not neutralizing. These antibodies are widely used as **serological biomarkers** to assess previous infection or vaccine-induced immunity in patients, but they are **not direct therapeutic targets**. The spike and nucleocapsid proteins themselves are the canonical viral targets for immune responses and for some COVID-19 drugs and vaccines[1][2][3][4][5][6][7][9].

02

Mechanism of action

Neutralization of viral particles (anti-spike IgG blocks viral entry into host cells)[3][5][7][9] Opsonization and elimination of virus-infected cells (general role of anti-viral IgG) Diagnostic detection (used in serological assays)

03

Biological functions

Immune responseVirus neutralization (for anti-spike IgG)Diagnostic biomarker (serology)
04

Disease associations

Infection (specifically COVID-19)
05

Safety considerations

Potential for cross-reactivity with other coronaviruses (false positives in serology)Does not indicate current infection or protect all individuals equally (diagnostic limitations)
06

Biomarkers

Presence of IgG against spike protein: marker of prior infection or vaccination[3][5][9]Presence of IgG against nucleocapsid protein: marker of prior infection (not induced by spike-protein-only vaccines)[2][4][6]
