Overview

Immunoglobulin G4 (IgG4) specific for Bet v 1 is a specialized antibody subclass produced by the human immune system in response to exposure to the major birch pollen allergen, Bet v 1 (Shamji & Durham, 2017). These antibodies are primarily characterized by their role as blocking antibodies in the context of allergic diseases and allergen immunotherapy (AIT) (Akdis & Akdis, 2014). By binding to the same epitopes on the Bet v 1 protein as IgE antibodies, IgG4 competitively inhibits the formation of allergen-IgE complexes (Flicker et al., 2002). This inhibition prevents the cross-linking of high-affinity IgE receptors (FcεRI) on the surface of mast cells and basophils, thereby blocking the release of inflammatory mediators such as histamine. Furthermore, Bet v 1-specific IgG4 can interfere with IgE-facilitated antigen presentation to T cells, which helps shift the immune response from a pro-allergic Th2 profile toward a more tolerant state (Nouri-Aria et al., 2004). The induction of high levels of these antibodies is a key immunological marker of successful desensitization during birch pollen AIT. While naturally occurring as a result of immune modulation, recombinant versions of these antibodies are being researched as potential passive immunotherapy treatments to provide immediate protection against birch pollen-induced allergic rhinitis and asthma (Valenta et al., 2016).
