Overview

Immunoglobulin gamma Fc receptor I, commonly known as CD64 or FCGR1A, is a high-affinity receptor for the Fc region of immunoglobulin G (IgG). It plays a crucial role in both innate and adaptive immune responses by mediating various effector functions upon binding to IgG-coated targets such as pathogens or immune complexes. CD64 is constitutively expressed on monocytes and macrophages, and inducible on neutrophils and eosinophils during inflammatory conditions. Upon ligand binding (i.e., cross-linking by IgG), CD64 transduces signals through ITAM motifs present on its associated γ-chain, leading to cell activation including phagocytosis or cytokine production. Targeting CD64 may enhance or suppress immune responses depending on context, with potential applications in improving monoclonal antibody therapies or treating autoimmune diseases.
