Overview

Immunoglobulin heavy constant delta (IGHD) encodes the constant region of the delta heavy chain of immunoglobulin D (IgD), a class of antibodies primarily found as membrane-bound receptors on the surface of mature, naïve B cells, where it is co-expressed with IgM[1][3][5][11]. IgD molecules play a role in B cell activation and antigen recognition during the humoral immune response; the variable region contributes to specific antigen binding, while the constant delta region is involved in signaling intracellular activation after antigen encounter[3]. IgD can induce cytokine release from monocytes and is involved in both the recognition and effector phases of immunity, though its role is less prominent than that of IgM, IgG, or IgA[3][11]. It is not considered a therapeutic target in drug development but serves as an important marker in hematopathology and immunology. - IGHD is located on chromosome 14 and is highly conserved. - Diseases associated with IGHD include several lymphoid malignancies, primarily as part of immunophenotyping panels, not as a direct therapeutic target[1][6]. - IgD expression is regulated by cytokines (e.g., IL-27) and ceases following antigen encounter during B cell differentiation[3].

Mechanism of action

Not applicable (no drugs directly target the constant region of IgD)[3]. Most B cell-targeted therapeutics act by depleting cells expressing IgD, not by modifying the IGHD protein itself.

Biological functions

Immune responseAntigen bindingRegulation of cytokine production (e.g., TNF, IL1B, IL1RN, IL6, IL10, LIF)[1]Activation of B lymphocytesEffector function of humoral immunity[1][8][11]
04

Disease associations

Leukemia (Acute lymphoblastic)[1]Appendix lymphoma[1](Broadly, altered immunoglobulin D may be implicated in immune system dysfunction, though it is not a direct drug target.)
05

Safety considerations

None specifically documented for IGHD as a therapeutic target.Since IgD is not a drug target, there are no direct therapeutic safety concerns; therapies that deplete B cells may impact IgD-positive cells as part of the overall population.
06

Interacting drugs

None known (IgD is not commonly directly targeted by drugs)[1][3].

07

Biomarkers

IgD expression can serve as a marker for mature/naive B cells[3].Its presence may help in immunophenotyping of B cell populations in lymphoma/leukemia diagnosis[3].
