Immunoglobulin heavy constant gamma 1 (IGHG1) encodes the constant region of the heavy chain of IgG1, the most prevalent subclass of IgG antibodies in human serum[1][4][7]. The IgG1 molecule is comprised of two heavy chains (with IGHG1 as their constant region component) and two light chains, forming a basic Y-shaped configuration critical for specific antigen recognition (via variable domains) and immune effector functions (via the Fc domain)[5][8][9]. IGHG1 enables the antibody's interaction with Fc gamma receptors (FcγR) and complement proteins, mediating antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity[1][2][4]. IgG1-based therapeutic monoclonal antibodies use this domain as the protein scaffold due to its potent effector functions; however, IGHG1 is not a direct drug target itself, but an essential building block for antibody-based drugs[8]. Altered expression or structure of IGHG1 can be associated with autoimmune disease, infection susceptibility, and various immune pathologies[1][4][8].
