Immunoglobulin heavy diversity 2-21 (IGHD2-21) is one of several diversity (D) gene segments located in the immunoglobulin heavy chain locus on chromosome 14 (14q32.33)[2][4]. During B cell development, IGHD2-21 can be recombined with variable (V) and joining (J) gene segments to form the variable region of the immunoglobulin heavy chain, a critical component of the antigen-binding site of antibodies[2]. This V(D)J recombination process underlies the immense diversity of the adaptive immune response, enabling the recognition of a vast array of antigens. Polymorphisms and structural variants in the IGH locus, including diversity segments like IGHD2-21, can influence the composition and function of an individual's antibody repertoire, affecting susceptibility to infections, responsiveness to vaccines, and potentially autoimmune diseases[3]. However, IGHD2-21 is not a stand-alone therapeutic target, receptor, or enzyme, but rather a genetic building block for antibody generation.
