Overview

Immunoglobulin heavy joining 3 (IGHJ3) is one of several *joining* (J) gene segments located in the immunoglobulin heavy-chain locus (IGH) on human chromosome 14q32.2. During B-cell development, IGHJ3 undergoes site-specific DNA recombination as part of V(D)J rearrangement, where one V (variable), one D (diversity), and one J (joining) segment are joined to create the variable region of the immunoglobulin (antibody) heavy chain. This process generates antibody diversity crucial for adaptive immunity. While IGHJ3 itself does not function as a protein, its DNA sequence is incorporated into the B-cell receptor or antibody molecule, contributing to antigen recognition and immune response. Rearranged IGH clonotypes, including those involving IGHJ3, are valuable biomarkers for identifying and tracking B-cell malignancies and responses to therapy. IGHJ3 is a germline gene segment, not a functional protein or receptor. Its primary role is in antibody gene recombination and it is utilized in diagnostic and research contexts, rather than as a direct drug target.
