Overview

Immunoglobulin heavy variable 3-23 (IGHV3-23) is one of approximately 40 functional heavy chain variable (V) genes found within the immunoglobulin heavy chain locus on chromosome 14[2][3][5]. It encodes the variable region of immunoglobulin heavy chains, which are key components of the antigen-binding site of antibodies[1][2][3]. IGHV3-23 is frequently over-represented among the human antibody repertoire and is involved in antigen recognition as part of both membrane-bound immunoglobulin B cell receptors and secreted antibodies[1][2][5]. The gene undergoes V-(D)-J recombination, followed by somatic hypermutation and affinity maturation, enabling the generation of high-affinity, diverse antibodies in response to a wide array of antigens, including self- and foreign molecules[1][5]. IGHV3-23 germline variants and polymorphisms can affect the diversity and characteristics of expressed antibodies, which in turn can influence susceptibility to diseases, variability in vaccine response, and autoimmunity[4][5]. IGHV3-23 itself is not a conventional drug therapeutic target (like a receptor or enzyme), but its usage and genetic variation are important in the context of immune monitoring, diagnostics, and understanding humoral immune responses[4][5].
