Overview

Immunoglobulin heavy variable 3-35 (IGHV3-35) is a probable non-functional open reading frame within the immunoglobulin heavy chain variable region locus, situated on chromosome 14q32.33. The IGHV3-35 gene segment encodes a variable domain that normally helps form the antigen binding site of immunoglobulins (antibodies); however, due to mutations, recombination defects, and/or conserved amino acid changes, IGHV3-35 is generally predicted to be non-functional and does not contribute to the synthesis of productive immunoglobulin heavy chains. Immunoglobulin heavy variable genes collectively enable the generation of antibody diversity via V-(D)-J recombination and somatic hypermutation, forming critical components of the adaptive immune response. IGHV allelic variation influences the structure and function of the antibody repertoire in the population, but the IGHV3-35 locus is typically absent or non-expressed in most individuals, so it does not participate in functional immune processes nor present as a drug target or biomarker.
