Immunoglobulin heavy variable 3-50 (IGHV3-50)

Molecular classification
Immunoglobulin gene segment, Variable region gene, Other (part of adaptive immune gene family)

Overview

Immunoglobulin heavy variable 3-50 (IGHV3-50) is a gene segment in the immunoglobulin heavy-chain locus encoding part of the variable region of antibody heavy chains in humans. It is used during V(D)J recombination in B cell development to create a diverse repertoire of antibodies, allowing the immune system to recognize a broad array of antigens[1][2][4]. IGHV3-50 is one of many highly polymorphic gene segments contributing to antibody diversity; its usage, mutation status, and allelic variants are relevant in immunology and as biomarkers in certain hematological malignancies but it is not a classic druggable target or immune receptor in itself.

Mechanism of action

Not applicable as a target. The gene segment encodes variable regions of antibodies; these antibodies can be engineered as therapeutics but IGHV3-50 itself is not modulated by drugs.

03

Biological functions

Antigen binding (via encoding antibody variable region)Generation of antibody diversity through V(D)J recombinationImmune response (adaptive immunity)
04

Disease associations

Cancer (especially as a biomarker in B cell malignancies, e.g. lymphomas, leukemias, where IGHV gene usage or mutation status is relevant[6])Infection (through involvement in antibody responses)Other (as a gene segment, not a direct disease driver, but its sequence usage can be related to disease prognosis)
05

Biomarkers

IGHV gene mutation status is a prognostic biomarker in B cell malignancies, such as chronic lymphocytic leukemia (CLL)[6].Specific IGHV gene segment usage can be tracked in minimal residual disease assays, immune repertoire analysis, or clonality assessments.
