Immunoglobulin heavy variable 3-66 (IGHV3-66) is a gene segment encoding the variable region of the immunoglobulin heavy chain, crucial for the diversity of antibody specificity. It is one of ~129 variable gene segments in the IGH locus on chromosome 14 and undergoes V(D)J recombination during B-cell development, forming part of the antigen-binding site in immunoglobulins. IGHV3-66 is expressed in B-lymphocytes, and the encoded protein participates in antigen recognition, forming the structural basis for the adaptive immune response. Polymorphisms in IGHV3-66 and allelic usage affect antibody diversity and can influence susceptibility to immune-mediated diseases such as Kawasaki disease. While not a direct drug target, IGHV3-66’s encoded antibodies (e.g. in monoclonal antibody therapies) may be relevant in infectious disease treatment and immune diagnostics.
