Overview

The immunoglobulin heavy variable 4-34 (IGHV4-34) gene encodes a variable region of the immunoglobulin heavy chain, a critical component of the B cell receptor (BCR) on B lymphocytes. This gene segment is notable for its intrinsic self-reactivity, particularly against I/i carbohydrate antigens, and its involvement in both protective immunity and autoimmune disease. While normally tightly regulated to prevent autoreactivity, dysregulation of IGHV4-34 expressing B cells can contribute to the pathogenesis of diseases like SLE and cold agglutinin disease. It also plays a role in certain B cell malignancies like hairy cell leukemia and CLL.
