Overview

Immunoglobulin heavy variable 4-61 (IGHV4-61) is a variable (V) gene segment within the immunoglobulin heavy chain (IGH) locus on human chromosome 14q32.33[1][6]. It encodes the V region of the heavy chain in antibodies, which forms part of the antigen binding site together with the light chain variable region[5][6][3]. As a variable gene segment, IGHV4-61 undergoes somatic recombination with diversity (D) and joining (J) gene segments during B-cell development, enabling the immune system to generate a vast diversity of antibodies for recognition of pathogens[1][2][5]. IGHV4-61 is not itself a druggable therapeutic target (such as a receptor or enzyme), but allelic variation in IGHV4-61, such as the IGHV4-61*02 allele, has been associated with increased risk of specific autoimmune conditions, notably rheumatic heart disease[2]. IGHV4-61 is significant as a component of the antibody repertoire and as a potential biomarker for disease risk, but it is not a receptor or traditional drug target in therapeutic development[6][2][5].
