Overview

Immunoglobulin kappa variable 3-20 (IGKV3-20) is a gene segment that encodes the variable (V) region of the kappa light chain in immunoglobulin molecules. The V region is a critical component in the antigen-binding site of antibodies, generated through V-J recombination in B cells, allowing for a diverse adaptive immune response[3][5]. IGKV3-20 is expressed in B cells and contributes to the specificity and affinity of antibody–antigen recognition[4][5][9]. The product of this gene is often used in the assembly of stereotypic immunoglobulin kappa light chains, particularly seen in conditions such as rheumatoid factor-positive autoimmune diseases and certain B cell malignancies[2][5][1]. IGKV3-20, especially when paired with specific heavy chain variable genes (e.g., IGHV1-69), has been implicated in the production of autoantibodies such as rheumatoid factors—antibodies directed against the Fc portion of IgG and associated with rheumatoid arthritis, cryoglobulinemia, and B cell lymphoproliferative disorders[2][1]. It does not qualify as a classic “therapeutic target” like a receptor, enzyme, or transporter, but is rather a molecular segment frequently studied for its role in antibody diversity, autoimmunity, and as a marker for certain disease states[5][9][1]. No direct drug-targeting interaction is recorded; the significance of IGKV3-20 lies in its participation in antigen binding and its association with disease-relevant antibody profiles.
