Immunoglobulin lambda-like polypeptide 1 (IGLL1) is a surrogate light chain component critical for early B-cell development, expressed primarily in pro-B and pre-B cells as part of the pre-B cell receptor complex. The protein enables B-cell differentiation, allelic exclusion of the immunoglobulin heavy chain, and facilitates immunoglobulin light chain gene rearrangements, all necessary steps for effective antibody production. IGLL1’s gene does not undergo immunoglobulin rearrangement, distinguishing it from typical immunoglobulin genes. Mutations in IGLL1 can cause autosomal recessive agammaglobulinemia, a severe B-cell deficiency resulting in very low antibody levels and immunodeficiency. Increased IGLL1 expression is associated with several B-cell malignancies, suggesting a possible biomarker role in leukemia and lymphoma diagnosis or prognosis. It belongs to the immunoglobulin gene superfamily and is membrane-bound in association with the pre-B cell receptor during early B-cell maturation.
