Immunoglobulin lambda-like polypeptide 5 (IGLL5) is a member of the immunoglobulin superfamily, encoded by the IGLL5 gene and located within the immunoglobulin lambda locus on human chromosome 22. Unlike classical immunoglobulin genes, IGLL5 does not require somatic rearrangement for its expression; its first exon is unrelated to immunoglobulin variable genes, while the second and third exons correspond to joining and constant segments. It participates in the immune response through antigen binding, but is not a component of assembled functional antibodies in the way classical V(D)J rearranged immunoglobulins are. IGLL5 is notable for its frequent mutation in chronic lymphocytic leukemia, where its mutational spectrum reflects aberrant activation-induced cytidine deaminase (AID) activity, suggesting a role in lymphoma pathogenesis as a mutational hotspot—however, it is not a therapeutic target. IGLL5 has multiple transcript variants due to alternative splicing.
