Immunoglobulin lambda variable 5-48 (IGLV5-48) encodes a probable non-functional variable region gene segment of the immunoglobulin lambda light chain. As part of the antibody variable region, it would normally contribute to antigen recognition in humoral immunity. The gene is located on chromosome 22 (22q11.2) and is classified as non-functional because it carries sequence alterations, recombination or splicing defects, or conserved amino acid changes that prevent the production of a functional immunoglobulin chain[1][4][6]. Immunoglobulin variable regions undergo V(D)J recombination to generate binding diversity, but many such segments, including IGLV5-48, do not contribute to productive antibody chains due to these rarely productive rearrangements[1][3]. Immunoglobulins are glycoproteins composed of two heavy and two light chains, with the light chains existing as either kappa (κ) or lambda (λ)[3][5][7]. The IGLV5-48 gene is part of the lambda variable gene family, but unlike functional segments, it is not known to play a role in disease, immune regulation, or as a biomarker or drug target.
