Immunoglobulin-like domain-containing receptor 2 (ILDR2) is a type I transmembrane protein that functions as a member of the B7 family of immunomodulatory receptors. It is involved in the negative regulation of T-cell activation, contributing to immune tolerance. ILDR2 is expressed in several non-lymphoid tissues, fibroblastic reticular cells in lymph nodes, and is important for immune cell recruitment. Pharmacological targeting of ILDR2, for example with the antibody BAY 1905254, promotes T-cell activation and anti-tumor immunity, and has shown potential efficacy in cancer and autoimmune disease models. ILDR2 also participates in epithelial tight junction integrity and may influence inner ear development and function.
