Immunoglobulin-like transcript 7 (ILT7), also known as CD85g and LILRA4, is an inhibitory receptor selectively expressed on human plasmacytoid dendritic cells (pDCs). It negatively regulates innate immune responses by suppressing pDC activation and type I interferon (IFN-I) production following stimulation through Toll-like receptors TLR7/9. The natural ligand for ILT7 is bone marrow stromal cell antigen 2 (BST2), also known as tetherin or CD317. Engagement between BST2 and ILT7 leads to suppression of IFN-I secretion by pDCs.
Engagement of ILT7 with its ligand BST2 leads to ITAM-mediated signaling via Src/Syk kinases through FcεRIγ, inhibiting pDC activation and IFN-I production.
