Overview

Immunoglobulin superfamily member 11 (IGSF11) is a cell-adhesion molecule belonging to the immunoglobulin superfamily, with high expression in the brain and testis. It mediates calcium-independent homophilic cell adhesion, regulates synaptic transmission and plasticity in neurons via interactions with PSD95, and maintains the blood-testis barrier for male fertility. IGSF11 is also quickly upregulated in osteoclast precursors, where it regulates osteoclast maturation and bone resorption through both cell-cell interaction and metabolic modulation (pyruvate kinase M2 activity). In immune cells, IGSF11 inhibits human T cell activation via its binding partner VSIG3. Aberrant expression is associated with diseases such as osteoarthritis and certain cancers, where it acts as a cancer/testis antigen and supports cancer cell growth.
