Immunoglobulin superfamily member 23 (IGSF23) is a transmembrane protein containing immunoglobulin-like domains, predominantly expressed in bone and small intestine, with particularly high levels in osteoclasts compared to osteoblasts and PBMCs. It localizes to the cell membrane and cytoplasm, not the nucleus. Functionally, IGSF23 has been implicated in osteoclast differentiation via MAPK signaling; a human loss-of-function mutation causes defective osteoclastogenesis and osteopetrosis, suggesting a key role in bone remodeling. While the broader immunoglobulin superfamily is crucial for immune signal transduction and cell adhesion, direct evidence for IGSF23’s involvement outside bone/osteoclast function is currently limited. IGSF23 is not currently recognized as a therapeutic target, biomarker, or drug interaction partner.
