Immunoglobulin superfamily member 3 (IGSF3) is a cell surface adhesion molecule from the immunoglobulin superfamily (IgSF), characterized by eight Ig-like domains and belonging to the EWI subfamily defined by the Glu-Trp-Ile (EWI) motif[2][4]. IGSF3 is a single-pass type I membrane protein highly expressed in placenta, kidney, lung, and the developing brain[2][4]. Its functions include regulation of neuronal morphogenesis, cell adhesion, migration, maintenance of epithelial barrier function, and modulation of cell cycle and proliferation, with particular relevance in cancer progression (hepatocellular carcinoma, glioma), wound repair, and possibly lung and neurodevelopmental disorders[1][3][4][9]. It interacts homophilically and with tetraspanins, impacts sphingolipid homeostasis, and may be a potential biomarker or therapeutic target for drug resistance (gemcitabine) in NSCLC[4][9][1]. Targeting IGSF3 has therapeutic potential but may pose safety concerns due to its developmental and tumor-promoting functions.
