Overview

Immunoglobulin superfamily member 6 (IGSF6) is a transmembrane protein belonging to the immunoglobulin superfamily, predominantly expressed on cells of the immune system such as dendritic cells, macrophages, white blood cells, and secondary lymphoid organs. It is predicted to enable transmembrane signaling receptor activity and has been implicated in cell surface receptor signaling pathways and immune response regulation. The gene is located on chromosome 16p12.2, entirely within an intron of the METTL9 gene. Its expression and gene network connectivity have been studied in host resistance to infectious challenges and inflammatory diseases, with roles in embryonic development and cell proliferation also suggested through temporally regulated expression in developing organs. Although initially identified in loci associated with inflammatory bowel disease, no direct association with disease susceptibility via common variants has been shown. It has been implicated in gene signatures linked to disease processes such as atrial fibrillation, multiple sclerosis, and cancer survival. Its precise molecular mechanisms and functions in pathologies remain to be fully elucidated, and no specific drugs target IGSF6 as of current knowledge.
