Target intelligence / Profile preview
Immunologic effector and regulatory pathways encompass the diverse biological processes used by the immune system to eliminate pathogens and maintain self-tolerance (Janeway et al., Immunobiology, 2001). Effector pathways involve the mobilization of cells like cytotoxic T-lymphocytes and the production of pro-inflammatory cytokines to neutralize threats (Abbas et al., Cellular and Molecular Immunology, 2021). Regulatory pathways, including those mediated by CTLA-4 and PD-1, act as checkpoints to prevent excessive inflammation and autoimmunity (Sharpe & Pauken, Nature Reviews Immunology, 2018). Dysregulation of these pathways is a hallmark of many diseases; for instance, cancer cells often exploit regulatory pathways to evade immune detection, while autoimmune diseases result from a failure of these same mechanisms (Waldmann, Nature Medicine, 2003). Pharmacological modulation of these pathways, such as through checkpoint inhibitors or TNF-alpha blockers, has revolutionized the treatment of oncology and chronic inflammatory conditions (Pardoll, Nature Reviews Cancer, 2012). Because this term refers to a broad set of biological mechanisms rather than a single molecular entity, it is classified as a pathway category rather than a specific therapeutic target.
Drugs targeting these pathways act through various mechanisms including checkpoint inhibition, cytokine neutralization, receptor antagonism, and immunosuppression to modulate immune activity.
