Overview

Dysregulation of these pathways is a hallmark of many diseases; for instance, cancer cells often exploit regulatory pathways to evade immune detection, while autoimmune diseases result from a failure of these same mechanisms (Waldmann, Nature Medicine, 2003). Pharmacological modulation of these pathways, such as through checkpoint inhibitors or TNF-alpha blockers, has revolutionized the treatment of oncology and chronic inflammatory conditions (Pardoll, Nature Reviews Cancer, 2012). Because this term refers to a broad set of biological mechanisms rather than a single molecular entity, it is classified as a pathway category rather than a specific therapeutic target.
