Overview

The immunoproteasome beta 1 inducible subunit (LMP2/PSMB9) is a catalytic subunit of the immunoproteasome, a specialized proteasome complex involved in generating peptides for MHC class I presentation. It is induced by inflammatory cytokines like IFN-γ and TNF-α, replacing the constitutive β1 subunit in newly formed proteasomes. This replacement alters substrate specificity, favoring the production of peptides with hydrophobic C termini suitable for MHC I binding, thereby enhancing CD8+ T cell epitope generation and adaptive immune surveillance. Inhibitors targeting β1i are under investigation for modulating immune responses in autoimmune diseases and cancer therapy.
