Overview

Immunoproteasome subunit LMP2 (β1i or PSMB9) is a catalytically active component of the immunoproteasome, a specialized proteasome complex crucial for immune function. It replaces the standard proteasomal subunit β1, conferring chymotrypsin-like activity and enhancing the generation of antigenic peptides for MHC class I presentation to CD8+ T cells. LMP2 is upregulated by IFNγ and is involved in NF-kB signaling. Altered expression or function is implicated in autoimmunity, cancer, neurodegeneration, and cardiovascular diseases, making it a potential therapeutic target. Selective inhibitors like UK-101 and dual LMP7/LMP2 inhibitors are under development.
