Overview

Immunostimulating Complex (ISCOM) adjuvants are particulate nanostructures composed of cholesterol, phospholipids, and saponins (such as Quil-A), designed to deliver antigens and stimulate robust cellular and humoral immune responses. They function by enhancing antigen presentation to APCs, promoting dendritic cell maturation, and inducing a cytokine environment conducive to both antibody and cytotoxic T cell (CTL) responses. ISCOM-type adjuvants have been used in veterinary and experimental human vaccines, with demonstrated utility against a variety of infectious agents and tumors. They differ from molecular therapeutic targets in that they are not single proteins or receptors, but structured complexes used to boost vaccine efficacy[1][2][3].

Mechanism of action

Enhances immunogenicity by: - Facilitating antigen uptake by antigen-presenting cells (APCs)[1][2] - Promoting maturation and IL-12 secretion by dendritic cells[2] - Fostering both humoral (antibody) and cellular (T cell, CTL) immunity, balancing Th1/Th2 responses[1][2][3] - Engaging inflammasome pathways (e.g., NLRP3/caspase-1)[2]

03

Biological functions

Immune response (enhances antigen presentation, stimulates B and T cell activation, promotes cytokine release and DC maturation)
04

Disease associations

Infection (used in vaccines against viral, bacterial, and parasitic diseases)Cancer (can generate cellular immune responses against tumors)
05

Safety considerations

Local inflammation due to cytokine/chemokine inductionSystemic immune activationSaponin component (Quil-A) sometimes causes toxicity in non-purified formulations
