Overview

Immunosuppressive cells that express NKG2D ligands (MICA, MICB, ULBP1-6) can interact with the NKG2D receptor on NK and T cells. This interaction is central to immune surveillance but can also play a role in immune regulation and evasion. Tumor cells, viral-infected cells, and immunosuppressive cells may shed soluble forms of these ligands (sNKG2DL), which can downregulate surface expression on tumor/targeted tissues. Transforming growth factor-beta (TGF‑β) further suppresses effector function. Therapeutic strategies include enhancing ligand expression for cancer therapy or blocking soluble ligand effects to restore antitumor immunity.
