Impact RWD domain protein (IMPACT) is a member of the RWD domain-containing protein family that modulates translational control—particularly in neuronal cells—by interacting with GCN2, a kinase involved in the cellular stress response to amino acid deprivation. While many RWD domain proteins are implicated in processes such as SUMOylation and protein–protein interactions, IMPACT is distinguished by its regulatory role in maintaining translation under certain stress conditions, preventing inappropriate activation of the GCN2 pathway, and thus supporting normal neuronal function[5]. The molecular structure includes the RWD domain, a fold commonly involved in protein–protein interaction, but it does not display enzymatic activity or ligand binding typical of classical therapeutic targets like receptors or enzymes[1][5].
