Target intelligence / Profile preview

Importin-9 (IPO9)

Target
IPO9
Molecular classification
Importin (β-type karyopherin), Nuclear transport receptor, Histone chaperone
01

Overview

Importin-9 is a β-karyopherin nuclear transport receptor that mediates the active import of actin monomers, histone H2A-H2B, selected transcription factors, and proteasome components into the nucleus. Importin-9 is critical for maintaining nuclear actin pools required for transcription and DNA repair, sequesters histones to prevent premature chromatin binding, and controls chromosome condensation and segregation during meiosis, especially in gametogenesis.

02

Mechanism of action

Nuclear import; acts as a transport receptor binding nuclear localization signals (NLS) on cargo proteins and mediates their translocation through the nuclear pore complex. Chaperone-like mechanism shields histone H2A-H2B from DNA and other histones during transport.

03

Biological functions

Protein nuclear import, particularly actin monomers and histone H2A-H2B dimersMaintenance of nuclear actin pools necessary for transcription and DNA repairChromosome condensation and segregation during meiosis (in gametes)Nuclear import of proteasome componentsNuclear import of selected transcription factors (e.g., Sox2, c-Jun)
04

Disease associations

Sterility due to meiotic defects (in Drosophila), relevant for germline developmentImplicated in cellular processes relevant to cancer biology (through transcriptional control and DNA repair)
05

Safety considerations

No specific therapeutic safety concerns, as IPO9 is not currently a direct drug target.Loss-of-function in Drosophila causes sterility and chromosomal defects, implying potential issues if similarly targeted in germline cells of other organisms.
