Overview

Inactivated poliovirus antigen type 1 is a purified, non-infectious component of the poliovirus vaccine, typically derived from the Mahoney strain. The virus is cultivated in cell cultures, such as Vero cells, and then chemically inactivated using formaldehyde to ensure it cannot replicate or cause disease while maintaining its structural integrity for immune recognition (CDC, 2023). Its primary biological function is to serve as an immunogen that triggers the host's adaptive immune system to generate protective neutralizing antibodies (WHO, 2022). These antibodies circulate in the blood and prevent the wild-type poliovirus from spreading to the central nervous system, thereby protecting against paralytic poliomyelitis (StatPearls, 2023). As a critical component of modern immunization schedules, it is frequently administered in combination vaccines alongside antigens for diphtheria, tetanus, and pertussis (FDA, 2023). Because the virus is completely inactivated, this antigen carries no risk of vaccine-associated paralytic poliomyelitis, making it the preferred choice for polio eradication efforts in many regions (NIH, 2021). It specifically targets the Type 1 poliovirus, which has historically been the most prevalent cause of wild poliovirus paralysis globally.
