Overview

Inactivated poliovirus type 3 capsid antigen is a fundamental immunogenic component of the inactivated poliovirus vaccine (IPV), typically derived from the Saukett strain of the virus (StatPearls). The antigen consists of the four structural proteins (VP1, VP2, VP3, and VP4) that constitute the viral capsid, which are treated with formaldehyde to abolish infectivity while maintaining the structural integrity required to elicit a protective immune response (UniProt, MicrobeNotes). Upon administration, these antigens are recognized by the host's immune system, triggering the production of serotype-specific neutralizing antibodies (PDR.net). These antibodies provide systemic protection by preventing the virus from spreading to the central nervous system, thereby averting the development of paralytic poliomyelitis (CDC). While IPV is exceptionally effective at preventing individual paralysis, it primarily induces humoral immunity and provides less robust mucosal (intestinal) immunity than live-attenuated vaccines (PolioEradication.org). Consequently, it is a vital tool in the global effort to eradicate polio, particularly in maintaining population immunity without the risk of vaccine-derived poliovirus transmission (Wikipedia).

Mechanism of action

The inactivated poliovirus type 3 capsid antigen induces active immunity by stimulating the production of serotype-specific neutralizing antibodies (IgG) in the serum (PDR.net, Drugs.com). These antibodies neutralize the virus during the viremic phase of infection, preventing its entry into the central nervous system and subsequent destruction of motor neurons (CDC, PolioEradication.org).
