Overview

The inducible caspase 9 (iCasp9) suicide switch is a genetically engineered safety mechanism designed to selectively induce apoptosis in modified cells, such as T cells used in adoptive cell therapies. This system provides a rapid and controllable way to eliminate therapeutic cells if severe adverse effects or off-target toxicities occur, thereby improving the safety profile of cellular immunotherapies. It consists of the intracellular domain of human caspase-9 fused to a modified drug-binding domain from human FKBP12. Activation requires a small-molecule CID like AP1903/Rimiducid, which dimerizes iCasp9, triggering apoptosis.
