Overview

Inducible nitric oxide synthase (iNOS or NOS2) is a key enzyme that produces high levels of nitric oxide (NO) in response to inflammatory stimuli, such as bacterial toxins or cytokines (UniProt P35228). Pro-inflammatory cytokines, including TNF-alpha, IL-1 beta, and IL-6, are small signaling proteins that orchestrate the body's immune response and amplify inflammatory signals (StatPearls PMID: 31855367). This collective group of molecules acts as a central driver in the pathogenesis of chronic inflammatory conditions, where their over-activation leads to tissue destruction and oxidative stress (PubMed PMID: 11592333). Therapeutic intervention typically targets these components individually, using small molecule inhibitors for iNOS or monoclonal antibodies to neutralize specific cytokines (PubChem CID 2146). While effective in reducing inflammation, targeting these pathways carries risks such as compromised host defense and increased susceptibility to opportunistic infections.
