Overview

Inducible T-cell co-stimulator ligand (ICOSL), also known as B7-H2 and CD275, is a cell surface protein and the primary ligand for the inducible T-cell co-stimulator (ICOS) receptor. It is expressed on antigen-presenting cells like B cells, macrophages, and dendritic cells. The ICOS/ICOSL interaction provides a crucial co-stimulatory signal for T-cell activation and function, regulating cytokine secretion, germinal center formation, and antibody class switching. ICOSL can also mediate reverse signaling in myeloid-derived antigen-presenting cells. Dysregulation of the ICOS/ICOSL pathway is implicated in autoimmunity, cancer, and other diseases, making it a therapeutic target.
