Overview

ICP34.5 is a multifunctional neurovirulence factor of herpes simplex virus type 1 (HSV-1). It plays essential roles in viral replication, evasion of host immune responses, and pathogenesis within the nervous system. It counteracts host antiviral responses by recruiting protein phosphatase 1 alpha (PP1α) to dephosphorylate eIF2α, reversing host-induced translational shutoff. It also interacts with Beclin 1 to inhibit autophagy. Loss or mutation in ICP34.5 leads to reduced neurovirulence, impaired replication, increased immune recognition, and attenuated pathogenicity.
