The NLRP3 inflammasome is a cytosolic multiprotein complex that plays a central role in the innate immune system by mediating inflammatory responses and cell death. It comprises the sensor NLRP3, the adaptor ASC, and the effector pro-caspase-1. Upon activation by diverse stimuli, NLRP3 oligomerizes, recruits ASC, and activates caspase-1, leading to the maturation and release of IL-1β and IL-18, and pyroptosis. Aberrant activation of the NLRP3 inflammasome contributes to various autoinflammatory, autoimmune, and other diseases, making it an important therapeutic target.
