The inflammasome pathway is a critical component of the innate immune system, acting as a first line of defense against pathogens and cellular stress. Inflammasomes are cytosolic multiprotein complexes that detect pathogen-associated molecular patterns (PAMPs) and danger-associated molecular patterns (DAMPs), leading to the activation of inflammatory responses and a specialized form of cell death called pyroptosis. Key components include sensor proteins (e.g., NLRP3, AIM2, NLRC4), the adaptor protein ASC, and the effector enzyme pro-caspase-1. Activation results in the cleavage and release of pro-inflammatory cytokines IL‑1β and IL‑18, as well as gasdermin D (GSDMD)-mediated pyroptosis. Dysregulation of the inflammasome pathway is implicated in various chronic inflammatory diseases, making it a promising therapeutic target.
