Overview

In a physiological context, these genes coordinate the immune system's reaction to tissue damage and pathogens, ensuring host survival through the recruitment of immune cells and the production of chemical mediators (Nature Reviews Immunology, 2011). However, the persistent or dysregulated expression of specific genes within this category, such as TNF, IL6, and PTGS2 (COX-2), is a primary driver in the pathology of chronic conditions including rheumatoid arthritis, inflammatory bowel disease, and atherosclerosis (NCBI, PMC3492709). Therapeutic strategies generally focus on the protein products of these genes; for example, monoclonal antibodies such as adalimumab and infliximab are used to neutralize TNF-alpha (PubMed, 22444342). Additionally, drugs like corticosteroids can broadly suppress the transcription of many pro-inflammatory genes to reduce systemic inflammation (Mayo Clinic, 2023). Because this term encompasses a wide range of molecular entities rather than a single receptor or enzyme, it is classified as a functional gene set or pathway rather than a discrete therapeutic target. Consequently, therapeutic modulation often requires a balance between reducing pathological inflammation and maintaining adequate immune surveillance against infections (NIH, 2022).
