Overview

Inflammation-responsive cardiovascular tissue refers to the pathological state of the heart and vasculature where inflammatory processes drive disease progression, such as in atherosclerosis or following a myocardial infarction (Libby, P., 2021, Nature Reviews Cardiology). These tissues are characterized by the recruitment of leukocytes and the secretion of pro-inflammatory cytokines like IL-1β and IL-6 (Nahrendorf, M., 2018, Nature Medicine). In drug discovery and bioengineering, this environment is treated as a therapeutic site where stimuli-responsive biomaterials can release drugs in response to local triggers like reactive oxygen species or matrix metalloproteinases (Fan et al., 2019, Advanced Drug Delivery Reviews). Clinical validation for targeting these tissues was provided by the CANTOS trial, which showed that inhibiting IL-1β reduces cardiovascular event rates (Ridker et al., 2017, NEJM). However, the primary challenge lies in modulating these tissues without causing systemic immunosuppression or disrupting the necessary healing phases of the heart (Swirski & Nahrendorf, 2013, Science). This approach represents a shift toward precision medicine by focusing on the specific inflammatory milieu of the diseased cardiovascular system.
