Overview

Inflammatory mediator signaling refers to the complex network of biochemical pathways through which cells communicate to initiate and regulate the inflammatory response. This process involves a wide array of signaling molecules, including cytokines (such as TNF-alpha and IL-6), chemokines, and lipid mediators like prostaglandins and leukotrienes (Nature Reviews Immunology, 2017). These mediators bind to specific cell-surface receptors, triggering intracellular cascades such as the NF-kappaB, MAPK, and JAK/STAT pathways, which ultimately lead to changes in gene expression and cellular behavior (StatPearls, 2023). While essential for host defense and tissue repair, dysregulated inflammatory signaling is a hallmark of numerous chronic conditions, including rheumatoid arthritis, inflammatory bowel disease, and atherosclerosis (PubMed, 2021). Consequently, many therapeutic strategies focus on modulating specific components of these pathways to alleviate symptoms and prevent tissue damage. However, because these pathways are also vital for normal immune function, systemic inhibition can lead to significant side effects, such as increased susceptibility to infections (NIH, 2022).
