Overview

Inflammatory pathways and mediators encompass a complex network of signaling cascades and biochemical substances that coordinate the body's response to injury, infection, or tissue stress [1, 7]. These pathways involve various cell types, including leukocytes, macrophages, and endothelial cells, which release mediators such as cytokines (e.g., TNF-alpha, IL-1, IL-6), chemokines, and lipid-derived eicosanoids like prostaglandins and leukotrienes [1, 6, 10]. While acute inflammation is a protective mechanism essential for healing and pathogen clearance, chronic or dysregulated activation of these pathways is a hallmark of numerous pathological conditions, including rheumatoid arthritis, inflammatory bowel disease, and atherosclerosis [7, 9, 13]. Therapeutic intervention typically focuses on specific components within these pathways, such as inhibiting cyclooxygenase enzymes with NSAIDs or neutralizing pro-inflammatory cytokines with monoclonal antibodies [1, 10, 18]. Understanding the interplay between these mediators is crucial for developing targeted therapies that can resolve inflammation without compromising the host's overall immune competence [15, 18].
