Inflammatory pathways in conjunctival goblet cells refer to the complex signaling networks, such as the JAK/STAT and NF-κB pathways, that regulate the survival and secretory function of these specialized cells (Pflugfelder et al., 2017, PMID: 28445318). Conjunctival goblet cells are essential for ocular surface health as they produce MUC5AC, a gel-forming mucin that stabilizes the tear film and protects the corneal epithelium (García-Posadas et al., 2016, PMID: 27101355). In chronic inflammatory conditions like Dry Eye Disease (DED) and Sjögren's syndrome, pro-inflammatory cytokines—specifically interferon-gamma (IFN-γ) and interleukin-17 (IL-17)—induce goblet cell apoptosis and dysfunction (Zhang et al., 2014, PMID: 25058338). Additionally, the activation of the NLRP3 inflammasome has been implicated in the inflammatory response within these cells (Chi et al., 2017, PMID: 28107479). This loss of goblet cells leads to mucin deficiency, tear film instability, and a self-perpetuating cycle of ocular surface damage. Therapeutic agents like Cyclosporine A and Lifitegrast are used to inhibit these inflammatory mediators, thereby preserving goblet cell density and restoring tear film integrity (De Paiva et al., 2016, PMID: 27554475). Monitoring goblet cell density and MUC5AC levels serves as a critical biomarker for assessing disease severity and treatment efficacy in clinical settings.
