Inflammatory signaling regulators encompass a diverse group of proteins, including cytokines, kinases, and transcription factors, that coordinate the complex biological response to harmful stimuli such as pathogens or damaged cells (StatPearls, 2023). Key pathways involved include the NF-κB, MAPK, and JAK-STAT cascades, which regulate the production of pro-inflammatory mediators like TNF-α and IL-6 (Nature Reviews Immunology, 2017). Dysregulation of these regulators is a hallmark of numerous pathologies, including rheumatoid arthritis, inflammatory bowel disease, and various cancers (PubMed, 2021). Therapeutic strategies often involve the use of monoclonal antibodies to neutralize cytokines or small molecules to inhibit intracellular kinases, thereby dampening the inflammatory response (NIH, 2022). However, because these regulators are also essential for normal immune surveillance, their pharmacological modulation can lead to significant safety concerns, most notably an increased risk of serious infections and potential malignancy (Frontiers in Pharmacology, 2020).
Inhibition of pro-inflammatory cytokines, antagonism of cell-surface receptors, and inhibition of intracellular signaling kinases (e.g., JAK, MAPK) to modulate gene expression and cellular responses (Nature Reviews Drug Discovery, 2018; PubMed, 2020).
