Overview

These vaccines utilize antigens from the A/California/07/2009 (H1N1)v-like virus, primarily the hemagglutinin and neuraminidase proteins, to induce active immunity (CDC, 2009). The mechanism involves the activation of B-cells to produce neutralizing antibodies and the priming of T-cells to recognize viral components (WHO, 2010). Clinical formulations included inactivated, adjuvanted, and live-attenuated vaccines such as Pandemrix and Focetria. While effective in controlling the pandemic, certain formulations were linked to rare safety concerns, most notably an increased incidence of narcolepsy associated with the AS03-adjuvanted Pandemrix vaccine in Europe (Taba et al., 2013; Partinen et al., 2012). Additionally, like other influenza vaccines, there was a monitored risk for Guillain-Barré syndrome (Salmon et al., 2013). Because the vaccine's efficacy depends on a complex biological cascade involving multiple cell types and signaling pathways, it does not possess a discrete protein target in the traditional pharmacological sense.
