Overview

The HA1 head domain of the Influenza A H5 hemagglutinin is a critical component of the viral surface glycoprotein responsible for initiating infection. It contains the receptor-binding site (RBS) that recognizes and attaches to alpha-2,3-linked sialic acids on host cell membranes, which is a hallmark of avian-adapted influenza viruses (UniProt: P03452). As the most immunodominant part of the virus, the HA1 head is the primary target for neutralizing antibodies elicited by infection or vaccination (PubMed: 28100607). However, this domain is subject to frequent antigenic drift, where mutations allow the virus to evade the host immune system, posing a significant challenge for vaccine development (NIH: PMC4776122). Therapeutic strategies targeting the H5 HA1 head include monoclonal antibodies and prepandemic vaccines designed to prevent zoonotic transmission and potential pandemic outbreaks (PubMed: 25855244).
