The Influenza A virus matrix segment mRNA, commonly referred to as Segment 7, is a polycistronic viral transcript essential for the replication cycle of the virus (Lamb & Lai, 1980). Through alternative splicing, this single mRNA segment encodes both the M1 matrix protein and the M2 ion channel protein (Palese & Shaw, 2007). The M1 protein provides structural support to the viral envelope and coordinates the assembly and budding of new virions, while the M2 protein acts as a proton-selective ion channel crucial for viral uncoating within host endosomes (Helenius, 1992). Because the sequences within Segment 7 are highly conserved across different influenza A subtypes, this mRNA is a primary target for novel antiviral strategies (Sui et al., 2009). Therapeutic approaches such as antisense oligonucleotides (ASOs), like AVI-7100, and small interfering RNAs (siRNAs) are designed to bind to specific sequences of the mRNA to trigger its degradation or block translation (Ge et al., 2003). By inhibiting the expression of both M1 and M2, these RNA-targeted drugs can effectively disrupt multiple stages of the viral life cycle, including entry, assembly, and release. This strategy is particularly valuable for addressing the challenge of drug-resistant strains that have rendered traditional M2-ion channel blockers like amantadine largely ineffective (Bright et al., 2006). Furthermore, targeting the mRNA allows for a broader spectrum of activity against various strains compared to targeting highly variable surface proteins like hemagglutinin.
