Overview

Influenza A virus subtype H5N1 is a highly pathogenic avian influenza (HPAI) virus belonging to the Orthomyxoviridae family. It primarily infects wild birds and poultry but poses a severe zoonotic threat to humans, often resulting in high mortality rates due to primary viral pneumonia and multi-organ failure. The virus possesses two key surface glycoproteins: Hemagglutinin (H5), which facilitates viral attachment to host cell sialic acid receptors and subsequent membrane fusion, and Neuraminidase (N1), which enables the release of progeny virions from the host cell surface. Pharmacological strategies against H5N1 focus on inhibiting these viral components; for instance, neuraminidase inhibitors like oseltamivir and zanamivir prevent viral spread, while baloxavir marboxil targets the viral polymerase complex. Due to its high mutation rate and the potential for genetic reassortment, H5N1 is a major focus of pandemic preparedness, necessitating constant surveillance and the development of both vaccines and broad-spectrum antivirals.
